Melanocytes, branched dendriticcells of neuro-ectodermal origin are specialized pigment producing cellslocated mostly at the base of the epidermis of vertebrate animal skin. Thepigment, melanin is synthesized from the amino acid tyrosine by tyrosinaseenzyme and stored in the special organelle called melanosomes as melaningranules. The melanin granules are transferred to surrounding keratinocytesthrough the melanocyte dendritic extremities and are responsible for skin,hair, and eye color in vertebrate organisms. Melanin pigment provides an effective protection to skin againstultraviolet radiation by absorbing the UV radiation and UV inducedintracellular free radicals. Our Melanocytes are derived from the human embryonicstem cells (hESC SEES3) using an in-house established protocol and speciallyformulated cell culture medium. The branched elaborated structures ofmelanocytes are clearly visible in the monolayer culture with or without matrixsupport. Melanocytes can grow and multiply in the Melanocyte Growth Medium (M254CF,Gibco Cat. no. M-254CF-500) supplemented with calcium plus Human Melanocyte Growth Supplement(HMGS, Gibco, HMGS, cat. # S-002-5). |
